COMPOSITION
Oncodox-10/50
Vial contains sterile mixture of

Doxorubicin HCL and lactose
• Description
• Doxorubicin is a cytotoxic anthracycline antibiotic
• Binds to nucleic acids with the double DNA helix
• Binds to cell membranes as well as plasma proteins

Dosage and administration
• On IV administration of doxorubicin ,extravasation may occur with or without an
   accompanying burning or stinging sensation
• If any sign of extravasation have occurred the injection or infusion should be
   immediately terminated and restarted in another vein
• If extravasation is suspected ,intermittent application of ice to the site for 15 min
   q.i.d for 3 daysmay be useful.
• The most commonly used dose schedule when used as a single agent is 60 to 75
   mg/m2 as a single iv injection administred at 21 days intervals
• When used in combination with other chemotherapy drugs,the most commonly
   used dosage of doxorubicin is 40 to 60 mg/m2 given as a single iv injection every
   21 to 28 days
• Doxorubicin dosage must be reduced in case of hyperbilirubinaemia as follows
• Plasma bilirubin dosage
• Concentration (mg/dl) reduction
• 1.2-3.0 50%
• 3.1-5.0 75%

Reconstitution directions
• Doxorubicin 10mg or 50mg vials should be reconstituted with 5ml and 25ml
   respectively of sterile water for injection to give a final concentration of 2mg/ml
   of doxorubicin HCL
• The rate of administration is dependent of size of the vein and the dosage
• The dosage should be administered in not less than 3 to 5 min

Contraindication
• Should not be started in pts with marked myelosupression induced by previous
   treatment with other antitumor agents or by radiotherapy
• Pts who received previous with complete cumulative doses of doxorubicin,
   daunorubicin,idarubicin and other anthracyclines

Warning and precautions
• Pregnancy
• The patient should be apprised of the potential hazard to the fetus
• Nursing mother
• Should be advised to discontinue nursing during doxorubicin therapy
• Side effects
• Cardiotoxicity
• Cutaneous-alopecia,hyperpigmentation of nailbeds,skin reaction
• GI-nausea,vomitting,stomatiits,oesophagitis
• Local-severe cellulitis,vesication and tissue necrosis
• Haematologic-acute myeloid leukemia
• Hypersensitivity
• Peripheral neurotoxicity
• Other- conjunctivitis and lacrimation

• Oncodox-10……..vial of 10 ml
• Oncodox-50……..vial of 50 ml
