Description
• Sertraline HCl is a selective serotonin reuptake inhibitor.
• It helps correct the chemical imbalance of serotonin in the brain.
• The effectiveness of Sertraline is presumed to be linked to its inhibition of CNS neuronal uptake of serotonin (5HT).
• Serotonin is a naturally occurring chemical in the brain that is involved in the transmission of messages between nerve cells.
• Sertraline hydrochloride is an antidepressant of the selective serotonin reuptake inhibitor (SSRI) class.• Sertraline shares the common side effects and contraindications with other members of SSRI class; however, it does not cause weight gain.
Selective Serotonin Reuptake Inhibitor –

SSRI for short - is a class of antidepressant drugs. These medications work by preventing the neurotransmitter serotonin from being reabsorbed by the nerve cell that released it, thus forcing the serotonin to remain actively working, which has been shown to ease depression in some patients.
• Taken orally, sertraline is absorbed slowly, achieving its maximal concentration in the plasma 4-6 hours after ingestion.
• 98.5% of sertraline in the blood is bound to the plasma proteins.
• Its half-life in the body is 13-45 hours and is about 1.5 times longer in women (32 hours) than in men (22 hours), resulting in the proportionally 1.5 higher exposure of women.
• Sertraline is metabolized by several cytochrome 450

Indications
Serdep is indicated in the treatment of

• Major Depressive Disorders
• Post-traumatic Stress Disorder (PTSD)
• Obsessive-Compulsive Disorder
• Panic Disorders

PTSD…… Events that might cause PTSD include rape, physical
abuse, an airplane or car crash, ...

Panic disorder……….. anxiety disorder & is characterized by unexpected
& repeated episodes of intense fear accompanied by chest pain, heart
palpitations, shortness of breath, dizziness, or abdominal distress.

 

Dosage
Major depression Adult Initially 50 mg daily. Obsessive-compulsive disorder Adult & adolescent 13-17 yr Initially 50 mg. Childern 6-12 yr Initially 25 mg. Panic disorder Initially 25 mg daily, may be increased to 50 mg once daily after 1 wk. If no clinical improvement is apparent, dosage may be increased at intervals of not less than 1 wk up to a max of 200 mg daily.
Administration
May be taken with or without food
Contraindications
Concomitant use w/ MAOIs.
Special Precautions
Seizures, mania or hypomania. Renal or hepatic impairment. Pregnancy & lactation.
Adverse Drug Reactions
Diarrhoea, vomiting, abdominal cramps, anxiety, agitation, nervousness, blurred vision, palpitation, hyponatraemia.

Drug Interactions
MAOIs, alcohol, lithium, IV diazepam, warfarin, digitoxin, tricyclic antidepressants.

Category C: Either studies in animals have revealed adverse effects on the foetus (teratogenic or embryocidal or other) and there are no controlled studies in women or studies in women and animals are not available. Drugs should be given only if the potential benefit justifies the potential risk to the foetus.
